The 3H3 monoclonal antibody reacts with mouse 4-1BB, a TNF receptor superfamily member also known as CD137. 4-1BB is a 39 kDa transmembrane protein expressed by T lymphocytes, NK cells, dendritic cells, granulocytes, and mast cells. 4-1BB (CD137, TNFRSF9) is a surface glycoprotein discovered by Kwon’s group as an activation antigen on the surface of T cells (Kwon and Weissman, 1989).Indeed, its regulated expression is induced upon CD3-TCR engagement and is further stimulated by CD28 ligation (Melero et al., 1998). The CD137 receptor plays a key role in mediating immune response by promoting T cell proliferation, survival, and memory. Effective agonism of CD137 has the potential to reinvigorate potent antitumor immunity either alone or in combination with other immune-checkpoint therapies. In this study, we describe the discovery and characterization of a unique CD137 agonist, 7A5, a fully human IgG1 Fc TNFRSF9 / 4-1BB / CD137 is a member of the TNF-receptor superfamily. This receptor contributes to the clonal expansion, survival, and development of T cells.

Expression of CD137 4-1BB on Human Follicular Dendritic Cells
On T and NK cells, CD137 is expressed following activation and, if ligated by its natural ligand (CD137L), conveys polyubiquitination-mediated signals via TNF receptor associated factor 2 that inhibit apoptosis, while enhancing proliferation and Se hela listan på academic.oup.com 4-1BB (CD137) is an activation-induced costimulatory molecule that is expressed on activated T cells, NK cells, dendritic cells, eosinophils, mast cells, endothelial cells, and some tumor cells. Anti-4-1BB mAs have demonstrated improved antitumor T-cell responses, with rejection of established syngeneic tumor cell lines in preclinical models.

CD137, also known as TNFRSF9 or 4-1BB, is an inducible costimulatory molecule expressed mainly on activated T cells. Its ligand, known as 4-1BBL, is expressed on activated macrophages, mature B cells, hematopoietic stem cells, and myeloid progenitor cells. Activated T cells also express CD137, and engagement with CD137 ligand enhances T cell proliferation, IL-2 secretion, survival, and cytotoxic activity. 4-1BB, also known as CD137 and TNFRSF9, is a transmembrane TNF receptor superfamily member that is expressed as on various populations of activated T cell including CD4+, CD8+, memory CD8+, NKT, and regulatory T cells as well as on myeloid and mast cell progenitors, dendritic cells, mast cells, and bacterially infected osteoblasts.

It is expressed on activated T cells. CD137 is a type I membrane protein and a member of the tumor necrosis factor receptor superfamily. CD137 appears to be important for T cell proliferation and survival, and induces monocyte activation through its interaction with 4-1BB ligand.
